概要
マラリア原虫はヒトの細胞(肝細胞と赤血球)に感染するステージと蚊のステージを行き来しながら生活環を築いています。特にヒト赤血球ステージでは無性生殖で継続的に増殖します。
一方、マラリア原虫は細胞内寄生原虫であるため、様々な代謝が単純化され寄生という生活環境に適応しています。細胞という構造体を形成する脂質代謝もその一つですが、マラリアにおける研究の歴史には創薬という大きなゴールが存在したため、基礎研究ではあまり注目されてきませんでした。
よってマラリア原虫の脂質代謝の研究は、哺乳動物における研究と比べて大きく遅れているのが現状です。現にヒトで同定されている脂質代謝関連酵素も、マラリア原虫における相同分子が見つかっていないものもたくさんあります。私たちは、細胞の形成や膜タンパク質の機能を支える「土台」としての脂質膜、エネルギーの貯蔵やシグナル分子として機能する多くの脂質分子に注目し、マラリア原虫の生物学の理解をいっそう深めたいと思っています。
From this nature of malaria study, lipid metabolism area is far behind that in mammalian studies. For example, many lipid metabolism-related enzymes that were well-studied in mammalian systems are not identified yet in malaria parasites. We are interested in lipid membranes which serve for constructing cell structures and supporting membrane proteins. We also interested in lipid molecules which store energy and function as signaling molecules for better understandings of malaria biology.主な研究アプローチは、脂質生化学、原虫の脂質代謝酵素の同定・遺伝子改変、生体膜の生物物理学的解析、蛍光イメージングであり、原虫の脂質代謝に重要な働きをしているタンパク質の機能と原虫のサバイバル機構の仕組みを探っています。
Our main study approaches include lipid biochemistry, genetic modifications of enzymes involved in lipid metabolism, biophysical analyses of biomembranes, and fluorescence microscopy analyses. With these cutting-edge technologies and knowledge, we study functions of proteins that support parasite’s lipid metabolism and survival tactics of malaria parasites.
