Trypanosoma cruzi was cultured with mouse fibroblasts as feeder layer cells. Amastigotes that were small and no flagella, proliferated in infected fibroblasts. Trypomasitgote had a long flagellum. In both stages nucleus and kinetoplast were stained heavily with Giemsa’s staining solution, and observed as large and small spot, respectively. Trypanosoma cruzi causes Chagas disease.
