 マラリア(Plasmodium falciparum)やシャーガス病(Trypanosoma cruzi)を引き起こす寄生原虫の生活環は、媒介昆虫内と宿主内のステージに大きく分けられ、各宿主内において生存するために形態・代謝を変化させ寄生環境に適応します。また、回虫の場合は成熟卵に汚染された食品を摂取する事で感染し、小腸で孵化したのち、幼虫が血流に侵入し、肺→胃と順に体内移行します。最終的には小腸で成虫となり虫卵が便とともに排泄されます。
 この様に、寄生虫(原虫・蠕虫)は個々の生活環において、中間宿主と終宿主または自由生活の各ステージにおける酸素濃度・温度・エネルギー源の違いと宿主の免疫反応といった、劇的な環境変化に対し巧妙に適応しつつ、「生き残る」メカニズムを保有し、進化してきました。この環境変化に速やかに適応するために寄生虫は柔軟な代謝経路を持つ必要があり、ミトコンドリアが極めて重要な役割を担います。
 例えば、回虫幼虫は体外で私たちと同じように酸素を用いた呼吸(酸素呼吸)を行うが、低酸素環境(小腸)で生育する回虫成虫は酸素の代わりにフマル酸を用いた呼吸(フマル酸呼吸)を行います。また、肝臓に寄生するエキノコックスの原頭節ステージや、腸管膜の門脈に寄生するマンソン住血吸虫の成虫では、酸素呼吸とフマル酸呼吸の両方を行える事を最近明らかにしました。興味深い事に、ガン微小環境下(低酸素・低栄養)で増殖する一部の固形ガンでは、回虫と類似したフマル酸呼吸を行っている事を明らかにしつつあります。近年、マラリア原虫の有性生殖期・蚊内期・肝内期においては、寄生虫の細胞分化・生存はミトコンドリアに強く依存する事が判ってきました。そのため、蚊内期を用いた実験系各種の確立を行っております。
 私たちのグループでは「ミトコンドリア」、「エネルギー代謝」、「環境適応」といったキーワードを基盤とし、寄生虫・細菌・ガンを対象に研究を進めています。研究アプローチとしては生化学、分子生物学、構造生物学、ケミカルバイオロジー、オミックス解析、創薬科学の手法を駆使し、将来的には分子創薬が期待できる基礎/応用研究を行っています。本分野ではマラリア原虫を主な研究対象としていますが、他の病原体やモデル生物を用いた研究も行っています。
 具体的にはアイメリア(Eimeria tenella)、タイレリア(Theileria spp.)トリパノソーマ(T. cruzi, T. brucei, Leishmania spp.)といった寄生原虫の他、ブタ回虫(Ascaris suum)、アニサキス(Anisakis spp.)、捻転胃虫(Haemonchus contortus)、肝蛭(Fasciola spp.)等の蠕虫、そして、細菌では結核菌(Mycobacterium spp.)、ピロリ菌(Helicobacter pylori)、カンピロバクター(Campylobacter jejuni)や大腸菌(Escherichia coli)について研究を行っています。

 Mitochondrion is an organelle known to be “cellular power plant” responsible to provide energy to sustain life. However, in addition to its key role on energy metabolism, it has several functions such as signaling, cellular differentiation, cell death and provides essential metabolites for other metabolic pathways. Importantly, most of those processes are depending on oxygen through a process called oxygen respiration. Most parasites have a life cycle alternating between vector/free living and host stages. Those environments are completely distinct, and thus parasites drastically change their energy metabolism in order to adapt to such environmental changes. Within the host, many parasites live under hypoxic environment with their mitochondria evolved in such a way where energy metabolism does not depend entirely on oxygen (Figure 1). Instead, it has an alternative respiratory pathway that uses fumarate (fumarate respiration) via CII (reverse reaction) or alternative oxidase leading to production of energy by CV or substrate level phosphorylation (Figure 1), although the exact molecular mechanisms are still unclear. The elucidation of parasitic mechanism of adaptation to hypoxic environment is important because it may reveal new insights to understand the parasitism phenomena and additionally, it may provide new target for drug and vaccine development.
 Our current research focus on elucidating the role of mitochondria in the mechanism of adaptation to hypoxic environment using variety source of organisms such as Helminths, Trypanosomatid, Apicomplexa and Dinoflagellate parasites. Our main collaborators in this field are Prof. Frederic Bringaud (University of Bordeaux), Prof. Mchael Boshart (University of Munich), Prof. Anthony Moore (Sussex University) and Prof. John Kelly (London School of Hygiene and Tropical Medicine). Once a drug target is biochemically characterized and validated, we determine its crystal structure, and identify potential inhibitors (Figure 2) either by high-throughput screening or by in silico screening. Promissing inhibitors are further studied through derivatization and chemical biology methods. We also develop several in vitro and in vivo evaluation tools to properly evaluate the derivatives synthesized by our collegues. In addition to target-based screening described above, we also conduct robust cell-based screening and attempt to identify the molecular taget of the hits by combination of multi-omics, generation and analysis of resistant parasite and genetic approaches.
